Annotation	In this special issue, some of the current theories and aetiologies of Alzheimer's disease (AD) are discussed. Adams opens the issue with an article that critically reviews and evaluates diverse hypotheses for AD in light of recent findings; three hypothetical models for disease progression are presented. Other articles present work by distinguished international scientists, who are working toward understanding causes of AD, so that strategies can be designed to prevent or cure this debilitating disease.
