Three-year changes in cognitive performance as a function of Apolipoprotein E genotype: evidence from very old adults without dementia

AnnotationResearch suggests that the presence of the e-4 variant of apolipoprotein (APOE) is associated with an increasing risk of developing Alzheimer's disease. This Swedish study examined whether baseline cognitive performance and 3-year longitudinal changes were influenced by APOE-e4 status. Participants consisted of 20 APOE-e4 and 54 non-APOE e-4 older adults aged 80 years and over without dementia participating in a population-based longitudinal study of ageing and dementia. Cognitive performance was indexed by the Mini-Mental State Examination (MMSE) and multiple indexes of memory, visuo-spatial, and verbal performance. The results indicated no significant baseline differences between the two groups in any cognitive performance measures. However, analyses revealed that APOE-e4 groups experienced greater negative change in recognition memory for faces and words. Changes in tasks assessing other abilities did not vary as a function of APOE status. The study concluded that APOE-e4 status may not influence cognitive status in adults without dementia, and suggested that when such changes do occur, these may be related to impending dementia, rather than to the influence of the specific genotype on cognition in normal ageing.
