Annotation | One hundred years since Alzheimer's disease (AD) was first reported, this dementia is as contentious as ever, but there is intimation of therapeutic solutions being available in the next five years. Eminent contributors aim to present an accessible overview of the major areas of knowledge or controversy. Included are chapters on the ameloid protein beta (how and why it is formed, the neuropathological consequences of its production, and how they may be halted) and tau and its role in the disease process. The relevance of genetic mutations and polymorphisms are discussed, and disease-related neurochemical changes are described, particularly the molecular processes involved in cholinergic degeneration. Current and existing new therapeutic avenues and possible future directions are emphasised. (RH). |
