Familial influence on variation in age of onset and behavioural phenotype in Alzheimer's disease

AnnotationThe reasons for the clinical heterogeneity of Alzheimer's disease (AD) are unknown. This study aimed to determine the familial (genotypic) influence on phenomenology (phenotype) in AD. Affected sibling pairs with AD were assessed for a range of cognitive and non-cognitive symptoms. Significant familial effects on age of onset and mood state, and a relatively high pairwise concordance for agitation were found. The study concluded that substantial familial influence on age of onset, depression and agitation suggests that genotype does influence phenotype in AD. Establishing the molecular basis for this phenotypic variation may prove relevant to other neuropsychiatric disorders. (AKM).
